Four related processes
Absorption concerns entry from the administration site; distribution concerns movement into tissues; metabolism changes the molecule; elimination removes it or its products. Researchers use measured samples and models to describe these processes. Route and formulation are part of the result.

How to read a concentration curve
A peak concentration and the time to that peak describe one part of exposure. AUC summarizes exposure across time, while a terminal half-life describes a particular decline phase. Two formulations can have similar total exposure and different peaks, so no single number captures the whole profile.
What PK does not establish alone
Exposure data help design and interpret trials. They do not independently prove efficacy, tolerability or an appropriate regimen for a different population. Check whether a study measured the parent molecule or a metabolite, and whether its participants match the clinical question.
